Mixer does not support the EnvyHT24 cards like the one sold by Acube.
But you still seem to have something wrong with your setup.

Tampering with the envy files somehow left a "stain" in my system
preventing the Mixer from working properly (or at all).
Always thought that these kind of things can happen only in
Microsoft world...
Deleting Mixer.prefs does not help...

Make sure that you are not running and older copy of Mixer from another location.
You can replace Mixer and EnvyHT24 files with the ones from the AmigaOS4.1 install CD and reboot.
AmiUpdate should give you the option to update Mixer and the Envy24ht files.
If AmiUpdate does not it means that something is not right.

The only other place I can think of right now that might possibly "remember" anything about that wrong driver is the AHI prefs. You might want to try deleting your ahi.prefs file from envarc:sys (or move it elsewhere if you want to play it safe).

Although I doubt it, it's worth a quick shot.

P.S. My card is Envy24HT. But if I use files Envy24HT.audio and ENVY24HT in AHI,
Sam does not even start properly (no AmiDock, no RAWBinfo, etc...).
I may have missed it, but you never said what card you have, only what chip it uses.

Your problems does not sound like a software but a hardware problem, especially a card that is not seated right or, worse, that may either be not supported (hardware-wise) or broken in some way.

You told us that, if the card is in place the problems start, regardless if the drivers are installed or not, though the drivers in places make it worse.

What is the name of your card? (i.e. Terratec Aureon Space)

I'd suggest to get the card out, clean the contacts, reseat it and double check that it's firmly in place and try again without the drivers installed.
Now check all the problemmakers like "find" to work correctly and only then install the HT drivers and test further.

I don't know your hardware but the problem you described had the same effects on my XE (and it was a PCI card gone out of it's seating back then)

I bought the card from Acube Systems. Seems to me it has no real name (it is named
as Envy24HT PCIe sound card), but here are the specs:

The card is recognized by Ranger, so the physical connection is OK.

But what about this: The Mixer started to work properly (gui showing up, "Find" working, etc.) when I
removed the card and restored ahi.prefs from OS4.1 cd...and did the usual setup with Prefs:AHI for the
on-board sound. And after that putting the card back was enough to cause same problems as before.
And when I took it out again those problems were gone...So, is the card somehow faulty?

You have to get the new version of the Envy24HT driver installed via Amiupdate before you make any conclusions .
Mixer should normally ignore the card so something may be confusing Mixer.
I have one of these cards in my Sam460ex too so I know what I am talking about.

You "could" have gotten a faulty card, that happens with the best companies.

Do as Spectre660 advised and get the latest Envy24HT driver from either AmiUpdate or from the depot.
EDIT: Seems like the latest version (5.17) is only available throgh AmiUpdate, but you could at least try to reinstall 5.12 first.

If you still got problems and not have the chance to try it out in another machine you might as well contact ACube for a repair/swap

This has nothing to do with the audio problem in hand, but If I may. I'd
like to handle another "irritating" issue concerning my AmigaOne500...

Is this normal?
When I start my amy with usb stick (having backup of WB) inserted, it won't
boot at all...it just turns off...a few seconds after the Uboot has printed
the text "Init USB...".

This is the normal behavior.
So you have to remove the usb stick when doing a cold boot .
